Description: Two independent assays were performed using different PAMPs: LPS (Escherichia coli serotype 0111: B4, Sigma-Aldrich), poly(I:C) (27-4729-01, Amersham Biosciences), Zymosan A from Saccharomyces cerevisiae (ZYM; Z4250, Sigma-Aldrich), PGN from Staphylococcus aureus (77140, Sigma-Aldrich) and LTA from Staphylococcus aureus (tlrl-slta, InvivoGen). In the first experiment, three rectangular 100 L tanks each containing 20 soles (17.8 ± 4.3 g) were prepared. Temperature (19 ºC) and salinity (36 ppt) remained stable throughout the experiments. After two days of acclimation, three experimental treatments were assayed in duplicate. Animals were anesthetized with phenoxyethanol (50 ppm) and then intraperitoneally injected with 100 µL of PBS, LPS (30 mg/Kg) and Poly(I:C) (15 mg/Kg). After injection, animal recovered normally without any stress signs. Six specimens for each treatment were euthanized (100 ppm) and sampled at 3, 12 and 24 h post inoculation (p.i.). Head kidney was rapidly dissected, frozen in liquid nitrogen and stored at -80ºC until use.In the second experiment, 18 soles were transferred to all eight 100L rectangular tanks. After five days of adaptation four experimental treatments were assayed in duplicate. In this case, animals were intraperitoneally injected with 100 µLof PBS, ZYM (40 mg/Kg), PGN (10 mg/Kg) and LTA (5 mg/Kg). Manipulation conditions and sample collection were as described above. Head kidney, spleen,brain, thymus and gills were rapidly dissected for further analysis, frozen in liquid nitrogen and stored at -80ºC until use.
